2000 Lexus RX300 150,000 miles. Started vehicle this AM, driving to work. Transmission wouldn't shift right. From drive it would shift 1-2-3 but would not shift out from 3-4. I manually shifted from 1(L)-2-3 would not shift from 3-4. Check engine light is blinking. Any ideas?

If the check engin e light is blinking, then you have a severe misfire issue. Get the codes read and go from there.
